The Effectiveness of Pimecrolimus in Airway Inflammation and Bronchial Hyperresponsiveness in Murine Asthma Model

Abstract

BACKGROUND
Pimecrolimus is effective in atopic dermatitis and psoriasis. However, little is known about the effectiveness of pimecrolimus in allergic asthma.
OBJECTIVE
The aim of this study was to assess the effectiveness of pimecrolimus in bronchial hyperresponsiveness and airway inflammation in murine asthma model. METHOD: Female BALB/c mice fed with a standard chow incorporated with pimecrolimus for one week before either sensitization (group A, n=10) or allergen challenge (group B, n=10). Enhanced pause (Penh), methacholine bronchoprovocation test and bronchoalveolar lavage fluid (BALF) analyses were performed. RESULT: The developments of airway narrowing and bronchial hyperresponsiveness induced by allergen challenge were suppressed in group B. The percentage of eosinophils and the levels of interleukin (IL)-4 and IL-5 in the BALF were also decreased in group B. CONCLUSION: Pimecrolimus given before allergen challenge suppresses the developments of airway inflammation and bronchial hyperresponsiveness in murine asthma model.
