The Anticancer Effect of Combined Intratumor Cisplatin Injection and Interstitial KTP Laser Therapy on Heterotransplanted Head and Neck Carcinoma

Abstract

BACKGROUND AND OBJECTIVES: Interstitial laser therapy (ILT)has cosmetic usefulness and is also important in preserving function, especially in head and neck cancer. Cisplatin is a key drug in chemotherapy for treatment of head and neck cancer. The current study was conducted to evaluate the synergistic effect of locally injected cisplatin-epigel and interstitial KTP laser therapy induced hyperthermia.
MATERIALS AND METHODS
SNU-1041 (10(7) cells/0.1 ml)cells were heterotransplanted to the back of a nude mouse by subcutaneous injection. ILT group (n=10)was treated by KTP laser 1 J/mm3 via a cylindrical diffuser tip inserted into the tumor with temperature monitored at 43-45degreesC. The combined treatment group (n=10)was treated by intratumor injection of 100-200 ug of cisplatin in a slow releasing, collagen-based gel carrier (Cisplatin-Epigel)4 hours before ILT to induce local hyperthermia. After 4 weeks of follow up, the treated tumors were evaluated by the cure rate and volume changes.
RESULTS
Eight of the combined treatment group (80%)showed complete remission of tumor at 4 weeks follow up, whereas only 3 (30%)in the ILT group showed remission (p<0.01 by chi-square test). SUMMARY: The current study showed the synergistic effect of combined local cisplatin injection and ILT induced hyperthermia on heterotransplanted human hypopharyngeal squamous cell carcinoma.
