Enzyme-Linked Immunosorbent Assay (ELISA) for the Detection of Antibody to Mycobacterium tuberculosis in Sera from Patients with Pulmonary Tuberculosis

Abstract

The leveIs of IgG antibody binding to the soIuble antigens of Mycobacterium boνis BCG in sera from patients with tuberculosis and healthy individuals in Korea were measured by the enzyme-Iinked immunosorbent assay (ELISA). The most reactive antigenic components with homologous goat hyperimmune serum distributed in the fraction, which was extracted from Iive BCG cells homogenized by French-press at 15, 000 psi and was precipitable at 75% saturation of ammonium sulfate. Thirty eight (79.2) out of 48 sputum positive patients and 23 (45.1 %) out of 51 sputum negative but radiologicaIIy suspected cases showed a positive reaction at the serum diIution 1 : 400, at which none of 24 healthy individuals showed a positive reaction. At a serum diIution 1: 3, 200, 29.2% of sputum positive patients showed a positive reaction whiIe 11.8% of sputum negative cases showed a positive reaction. Of 13 smear negative culture positive cases, 10 were positive at the serum diIution 1 : 400. Fifteen of 24 heaIthy individuals were positive at the serum diIution 1 : 50, 6 at 1 : 100, and 1 at 1 : 200.
