Evaluation of Slide and Tube Coagulase Tests, and Latex Agglutination Kit for the Identification of Staphylcoccus aureus

Abstract

BACKGROUND: Identification procedures of Staphylococcus aureus in our hospital were time-consuming and laborious because we simultaneously tested tube coagulase test using human citrated plasma, Dnase, and mannitol test for the confirmation of S. aureus. Furthermore, isolation rate of methicillin resistant S. aureus (MRSA) in recent years were about 70%. So, we evaluated five coagulase tests for the detection of accurate, rapid and simple identification procedure of S. aureus.
METHODS
A total of 165 Staphylococcus species isolated from clinical specimen of our hospital during July 1997 to Augest 1997 were tested. Identification of isolates were performed by conventional biochemical tests and commercial ID 32 STAPH (BioMerieux, SA, France). Coagulase tube and slide tests were performed by rabbit plasma and human citrated plasma, respectively. Staphaurex Plus were tested by manufacturer's instructions.
RESULTS
Sensitivity and negative predictive value of all five coagulase tests were 100%, and no isolates of 165 Staphylococcus species had false positive or false negative results on tube coagulase tests. The secificity and positive predictive value of slide coagulase tests, respectively, were as follows: rabbit, plasma slide test, 98.1 and 99.1%; Staphaurex Plus, 96.2 and 98.2%, and human plasma slide test, 54.7 and 82.4%. Three were no false positive or false negative results with five coagulase tests for 90 isolates of MRSA.
CONCLUSIONS
S. aureus was accurately identified by coagulase tube test using rabbit plasma and slide coagulase test using rabbit plasma was useful for the rapid identification of S. aureus.
