Immunohistochemical Study of S-100, HMB-45 and PCNA in Pigmented Nevus and Malignant Melanoma

Abstract
- The relative expression of S-100, HMB-45 and PCNA (proliferating cell nuclear antigen) was evaluated by immunohistochemical staining on the pigmented nevus and malignant melanoma. Tissue from 10 cases of melanoma, 10 cases of intradermal nevus, 7 cases of compound nevus and 8 cases of junctional nevus were studied to find out any informative clue for differential diagnosis. Reactivity was measured by intensity of staining with semi-quantitative scale (negative 0, weak +1, moderate +2, strong +3) in S-100, HMB-45, and PCNA positive cells were counted for percentile grade. the results were as follows; 1) S-100 was all positive in 35 cases of pigmentory lesions. 2) HMB-45 was selectively stained in malignant melanoma except spindle shape variant but few cases of compound and junctional nevi also showed positive cells of intraepidermal location. 3) PCNA positive cells were dominant in melanoma (mean 39+/-10.7%) with compare to benign pigmented nevi (below 1%).While interpretation of immunohistochemical staining based upon semiquantitative estimation, these results appear to have useful indication for predicting malignant changes in both HMB-45 and PCNA positive cases.
